《Coating Cantilever》

7:50

1. wash cantilevers by cold PBS three times
2. add avadin and put them in R.P. for 30 min
3. wash cantilevers by cold PBS twice
4. add con-A and put them in R.P. for 1 hour (due to analomy class)
5. wash cantilevers by cold PBS twice and add ddWater

《Collagen Coating》

10:00

1. Take dishes coated collagen out of the fridge
2. Wash by cold PBS twice
3. Add PBS and seal with parafilm

《Cell Subculture》

16:00

切完後細胞計數: 84 (1mL trypsin + 4mL DMEM)

1. 取1.5 mL加入10 cm dish
2. 取500μL離心，抽掉上清液加1500μL PBS，AFM用
3. (下次可以再少一點)

《單純coating collagen》

21:00

用一般UV照玻璃dish 20 分鐘

稀釋collagen(150μ collagen/3mL PBS)，加入玻璃dish，搖overnight

 

《換medium》

21:20

細胞有點滿(約8分)，但是因為明天要做實驗了所以沒有切，換medium而已

《AFM前製》

另一盤細胞因為長得很滿，怕太老，所以拿昨天切的細胞來切。

用DMEM終止trypsin反應後，離心，抽掉DMEM，加入PBS pipeting.

《AFM Experiment》

探針抓第一顆細胞時，沒有抓住，所以換了另一顆細胞

Gel上面有紅色sulpa-Sanpa的 紅點

想測試Coating Collagen後，照一般UV有何不同所以一盤有照UV15分鐘，另一盤(gel2)沒有照UV。

Washed by PBS once and then washed by DMEM three times.

Soak in DMEM, seal with parafilm and put them in the fridge.

• 切細胞，取1/3來做AFM實驗。
• 另外2/3繼續養在10 dish, P40→P41
• 細胞計數: 35 (3.5x10^5)

 

《AFM Experiment》

使用0號&5號針

此次的針可以使用，但跑出來的圖形怪怪的。

初步推測應是

1. collagen 沒有coating 好
2. 細胞不健康

《Coating Canteliver 3-2》

1. washed by cold PBS three times (fulfill the whole dish)
2. Add avadin and put it in room temperature
3. washed by cold PBS three times (fulfill the whole dish)
4. Add con-A and put it in room temperature. (At this step, the twin adhesive is not sticky anymore, so I change the twin adhesive again.)
5. Add dd water and seal with parafilm, store in the fridge.

Change medium.

Add DMEM and refridge in the fridge.

《Gel 2-2》

Time: 09:30

1. 照一般UV15分鐘
2. 倒掉collagen，washed by cold PBS
3. washed by DMEM three times
4. add DMEM and seal dishes with parafilm, store in the fridge.

《Coating Cantilever 2-2》

從ecubator中拿出來發現昨天忘記封parafilm了，所以biotin幾乎都蒸發掉了。

(不知道這樣會不會造成影響?)

1. Washed by iced PBS for 4 times (fullfill the whole dish twice)
2. Add avadin and put them in room temperature for 30 min.
3. Washed by iced PBS for 3 times (fulfill the whole dish once)
4. Add con-A and put them in room temperature for 30 min
5. Washed by iced PBS for 3 times (fulfill the whole dish once)

   (0號針在過程中斷掉了)

6. Add ddWater and seal the dish with parafilm, store in the fridge.

p.s. 四號針在乃哥整理冰箱的過程中不小心被打破了dish，所幸針沒有斷，所以再加滿dd Water，封parafilm冰在冰箱

《Cell》

今天下午看細胞時，發現細胞死掉了(細胞懸浮在medium中，都沒有貼附在dish上)

跟學長討論過後，再解凍細胞(液態氮存取編號4-1)

由於四號針又多了一個變數，所以乾脆再co一支新的針，五號針

18:00 加入Biotin-BSA, 封parafilm → ecubator

13:00 soak in 食人魚水

14:30  wash by ddWater several times, I filled up the whole dish by ddWater this time.

15:30 add biotin-BSA and put them into ecubator.

 

 

《Gel 2-1》

上次的膠有被污染的感覺，於是我重新過2mm，使用新的ddWater。

這次製作兩盤dish, 做膠前用酒精把dish洗乾淨，然後依著protocol做

放進冰箱shaker搖晃的時間為21:30，所以隔天早上九點來準備繼續做。

14:00 開始做膠，做到要加collagen前約16:00，泡在HEPES中等時間
16:00 針的食人魚水沖掉，wash by dd water for about 5 time and the add biotin, 封parafilm, 37度ecubator
17:30 切細胞，分兩盤
一號是要AFM的，二號繼續留著養


20:30 dilute collagen and add it into the dish,
21:00 put the dish on the shaker( in the 4 degree fridge)

今天做膠時DMEM incubate沒有在37度c而是在室溫(下次要在37度c)
co針最後一個步驟因為一個hiccup而毀了一切 Orz
以後做一盤要co兩支針，兩盤三支，以此類推
